<front><div type="abstract" xml:lang="en"><p>Oral health is characterized by functional oral polymorphonuclear neutrophils (<styled-content style="fixed-case">oPMN</styled-content>
s). Edentulism might be associated with a loss of <styled-content style="fixed-case">oPMN</styled-content>
s because these cells enter the oral cavity primarily through the gingival crevices. The main aim of this study was to investigate the numbers of <styled-content style="fixed-case">oPMN</styled-content>
s in rinse samples obtained from edentulous (<italic>n</italic>
= 21) and dentate (<italic>n</italic>
= 20) subjects. A second study aim was to investigate possible differences between <styled-content style="fixed-case">oPMN</styled-content>
s and peripheral blood polymorphonuclear neutrophils (<styled-content style="fixed-case">cPMN</styled-content>
s). Apoptosis/necrosis and cell‐activation markers (<styled-content style="fixed-case">CD</styled-content>
11b, <styled-content style="fixed-case">CD</styled-content>
63 and <styled-content style="fixed-case">CD</styled-content>
66b) were analyzed using flow cytometry. Reactive oxygen species (<styled-content style="fixed-case">ROS</styled-content>
) production was determined either without stimulation (constitutive) or in response to 10 <italic>μ</italic>
M phorbol myristate acetate or <italic>Fusobacterium nucleatum</italic>
. The edentulous subjects presented with lower <styled-content style="fixed-case">oPMN</styled-content>
counts and higher percentages of apoptotic/necrotic <styled-content style="fixed-case">oPMN</styled-content>
s compared with dentate subjects. Furthermore, <styled-content style="fixed-case">oPMN</styled-content>
s from edentulous donors expressed low levels of all three activation markers and low constitutive <styled-content style="fixed-case">ROS</styled-content>
. In contrast, <styled-content style="fixed-case">oPMN</styled-content>
s from dentate subjects expressed high levels of all three activation markers and a higher level of constitutive <styled-content style="fixed-case">ROS</styled-content>
than <styled-content style="fixed-case">cPMN</styled-content>
s. When challenged, <styled-content style="fixed-case">oPMN</styled-content>
s from edentulous subjects showed no upregulation in <styled-content style="fixed-case">ROS</styled-content>
production, whereas <styled-content style="fixed-case">oPMN</styled-content>
s from dentate subjects retained their ability to respond to stimulation. The functional characteristics of <styled-content style="fixed-case">cPMN</styled-content>
s were comparable between edentulous and dentate subjects. This study demonstrates that despite having functional <styled-content style="fixed-case">cPMN</styled-content>
s, edentulous subjects have low <styled-content style="fixed-case">oPMN</styled-content>
numbers that are functionally impaired.</p>
